Lysin with umbelliferyl glycosides
ATCV WAL lysin acting on a panel of umbelliferyl glycosides.
The substrate solution contained 25micrograms/ml methylumbelliferyl glycosides in distilled water. Assays contained 50microliters of substrate, 50microliters of buffer (25mM KOH, 50mM MES) and 2microliters of 100mM CaCl2. 10microliters ATCV WAL lysin was added except to the blank which contained the cellobioside. Assays were incubated overnight at RT and photographed with midrange UV illumination through a 520nm long pass filter.
The panel was observed at about 50min. At this time the N-Acetylglucosamidine appeared to be hydrolyzed to roughly twice the degree of the beta-1,4-glucoside.
