Chemical inhibition of PBCV lysin
Chemical inhibition of PBCV lysin
PBCV dissociated with 1M LiCl was treated with diethylpyrocarbonate (acetonitrile stock solutions, freshly prepared) for 15 min at 37C. 6ul aliquots were incubated with PBi cells (100ul) for 30 min at 37C. Triton X-100 was added and intact cells were pelleted.
PBCV dissociated with 1M LiCl was treated with DTT (aqueous stock solutions, freshly prepared) for 15 min at 37C. 6ul aliquots were incubated with PBi cells (100ul) for 30 min at 37C. Triton X-100 was added and intact cells were pelleted.
DEPC is an effective inhibitor. DTT appears to inhibit slightly at the highest (2mM)concentration
