Effect of ammonium sulfate on dsRNA extraction

CMV dsRNA from old tobacco leaves (very little RNA1+2); a batch of leaves was diced and mixed

Note that the pH 4.75 sodium acetate sample (far left) is badly contaminated

Increasing ammonium sulfate increases host RNA extraction. In general the more ammonium sulfate, the more host RNAs. The nature of these RNAs in unknown. Presumably they're small and have some ds character. Band sharpness suggests they're highly structured. Perhaps the faint cluster of bands near the bottom of the gel is tRNA.

Regardless of the nature of these RNAs it's clear that one can manipulate extractions by varying ammonium sulfate concentration.

Presumably the ammonium sulfate acts by increasing adsorption of RNAs to cellulose. If so one could compensate by reducing ethanol level.
