Effect of hydrogen peroxide on minipurification

Minipurifications with iodoacetamide or hydrogen peroxide

The samples are C. quinoa infected with "Rumex Virus", a nepovirus with a tripartite capsid (like cherry rasp leaf virus). In the middle lanes iodoacetamide is replaced by 3% hydrogen peroxide in the indicated amounts. Oxidation of DIECA to disulfuram (insoluble) shows that peroxide is active. With this tissue, peroxide treated samples have a slightly higher background. Surprisingly tissue extracts do not brown.
