Effect of pH on alginate lyase activity

1 microliter of Flavobacterium alginate lyase (20mg/ml) was added to 100 microliters of cells. The assay was incubated 24 hours at RT before adding Triton and centrifuging to remove cells.

All buffers are 25mM in K, pH 7 is MOPS, pH8 is Bicine, pH9 is AMPSO.



The pH optimum is obviously higher than that of phycodnavirus lysins.

Note that PBi is the only cell line susceptible to this lyase

Note also that the Flavobacterium lyase is suffciently dilute to mimimize autofluorescence.
