Screening commercial enzymes for lysin activity
Suspensions (100ul) of each of 4 cell types were incubated overnight at RT with mixtures of 3-4 commercial enzymes (20mg/ml, 5ul of each) in pH 6 MES buffer and then assayed for lysin activity (detergent mediated chlorophyll release.)
Set 1, Driselase, Hemicelluase (A. niger), Cellulase (A. niger), Glucuronidase (Glusulase)
Set 2, Cellulase RS (T. viride), Macerozyme, Xylanase (Thermomyces)
Set 3, Mureinase, Pectolyase, Chitinase (Brugia malayi)
Note the high (green) autofluorescence backgrounds. Set 1 enzymes have slight activity vs. PBi, Set 2 may have slight activity vs C169. Appropriate cell types will be tested vs. individual enzymes
